Yes, but only on one of the wheels.

Indeed you can, and lots of people have done so. The only drawback is that unless you have a fancy handle you won’t be able to look at it while you’re riding, as the computer will have to go on the seatpost.

Pay special attention to whereabouts on the wheel you mount the magnet. On a bike it doesn’t matter, but on a unicycle you want to ensure that the magnet doesn’t end up hovering next to the sensor while you’re hopping, idling or mounting; otherwise you may end up with massive maximum speeds before you’ve even gone anywhere…

And coker spoke are so fat that you might have to file the back of the magnet to fit it over the spoke. Then you won’t be able to put the screw in on the back, and so on and so on. So be carfull how you put it on.
